ABSTRACT

OBJECTIVE@#To explore the mechanisms of Buyang Huanwu Decoction (BYHWD) modulating the gut microbiome and trimethylamine oxide (TAMO) to exert cardioprotective effects.@*METHODS@#Ligation of the left anterior descending coronary artery was performed in rats to induce heart failure (HF). Except for the sham-operation group (n=10), 36 operation-induced models were randomized into 3 groups using a random number table (n=12 in each group): the model group, the BYHWD group (15.02 g/kg BYHWD), and the positive group (4.99 g/kg metoprolol succinate). After 4-week treatment (once daily by gavage), echocardiography was applied to evaluate the cardiac function and the Tei index (the ratio of ventricular isovolumic contraction time (IVCT) and isovolumic diastolic time (IVRT) to ejection time (ET)) was calculated; hematoxylin-eosin (HE) staining was observed to characterize the pathology of the myocardium and small intestinal villi. D-lactic acid was detected by an enzyme-linked immunosorbent assay (ELISA). Expressions of occludin, claudin-1, and zonula occludens (ZO-1) were detected by Western blot. 16S ribosomal ribonucleic acid (16S rRNA) sequencing was used to explore the changes in the intestinal flora. TMAO was detected via liquid chromatography-tandem mass spectrometry (LC-MS/MS).@*RESULTS@#In the echocardiography, the Tei index was considerably lower in the positive and BYHWD groups compared with the model group (P<0.05). Besides, BYHWD improved the pathology of myocardium and small intestine of HF rats and lowered the D-lactic acid content in the serum, when compared with the model group (P<0.05). BYHWD also improved the expression of occludin and claudin-1 (P<0.05); in the gut microbiota analysis, BYHWD slowed down modifications in the structure distribution of gut microbiota and regulated the diversity of intestinal flora in HF rats. The content of TMAO in the serum was significantly lowered by BYWHT compared with the model group (P<0.05).@*CONCLUSION@#BYHWD may delay progression of HF by enhancing the intestinal barrier structure, and regulating intestinal flora and TAMO.

ABSTRACT

Hepatic encephalopathy is a common metabolic neuropsychiatric syndrome in the development of end-stage liver disease. Since the concept of intestinal-liver-brain axis was proposed, the relationship between the pathogenesis of hepatic encephalopathy and the gut microbiota has been a hot research topic. In recent years, studies have confirmed that gut microbiota is involved in and affects various pathological processes of hepatic encephalopathy. This article combines the latest research progress at home and abroad to elaborate on the research status of regulating gut microbiota and thus interfering with the pathological process of hepatic encephalopathy, hoping to provide new ideas and methods for the intervention of hepatic encephalopathy based on the regulation of gut microbiota.

ABSTRACT

BACKGROUND@#The association of lipids and cancer has varied greatly among different cancer types, lipid components and study populations. This study is aimed to investigate the association of serum lipids and the risk of malignant lesions in esophageal squamous epithelium.@*METHODS@#In the "Endoscopic Screening for Esophageal Cancer in China" (ESECC) trial, serum samples were collected and tested for total cholesterol (TC), triglycerides, low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol at the time of subject enrollment. Cases were defined as malignant esophageal lesions identified by baseline endoscopic examination or by follow-up to May 31, 2018. Controls were randomly selected using incidence density sampling in the same cohort. Conditional logistic models were applied to identify the association of serum lipids and the risk of malignant esophageal lesions. Effect modification was evaluated by testing interaction terms of the factor under assessment and these serum lipid indicators.@*RESULTS@#No consistent association between serum lipid levels and esophageal malignant lesions were found in a pooled analysis of 211 cases and 2101 controls. For individuals with a family history of esophageal cancer (EC), high TC, and LDL-C were associated with a significantly increased risk of having malignant lesions (odds ratio [OR]High vs. Low TC = 2.22, 95% confidence interval [CI]: 1.14-4.35; ORHigh vs. Low LDL-C = 1.93, 95% CI: 1.01-3.65). However, a negative association was observed in participants without an EC family history (ORHigh vs. Low TC = 0.69, 95% CI: 0.48-0.98, Pinteraction = 0.002; ORHigh vs. Low LDL-C = 0.50, 95% CI: 0.34-0.76, Pinteraction < 0.001).@*CONCLUSIONS@#In this study, we found that the association of serum lipids and malignant esophageal lesions might be modified by EC family history. The stratified analysis would be crucial for population-based studies investigating the association of serum lipids and cancer. The mechanism by which a family history of EC modifies this association warrants further investigation.

ABSTRACT

Objective: To select the high altitude planting area for improving the quality of Angelica sinensis based on biomass, bioactive compounds accumulation and antioxidant capacity in rhizome. Methods: The experiments were conducted on dry weight (DW), bioactive compounds content and antioxidant capacity in two-year old rhizome of A. sinensis grown at different altitudes (2 100, 2 500, and 2 900 m), under controlling plants reproduced from the seeds of the same plant, the same levels of water and fertilizer in soil. Results: Higher altitude was adverse to rhizome biomass, however, the compounds content (DW and per plant basis) and antioxidant capacity were significantly promoted at higher altitude compared to lower altitude site; On a per plant basis, the content of ferulic acid, soluble sugar, phenolics and flavonoids in rhizome of un-bolted plants grown at 2 900 m respectively was 2.06, 1.13, 1.34 and 1.15 fold greater than that of 2 100 m; The antioxidant capacity also increased with higher altitude. Comprehensive analysis showed that total content of main bioactive compounds in rhizome of un-bolted plants increased with higher altitudes ranging from 2 100 to 2 900 m. Conclusion: Higher-altitude cultivation can significantly enhance bioactive compounds accumulation in rhizome. The biosynthesis and accumulation of bioactive compounds were regulated by low temperature and light. These findings will provide theoretical references for improving production and quality of rhizome as well as large-scale cultivation.

ABSTRACT

The dried roots of Angelica sinensis has been widely applied in clinical care due to its efficiencies in nourishing and activating blood, regulating female menstrual disorders and relieving pains, relaxing bowels, etc. The cultivated two-year-old plants normally harvested roots for medicinal uses emerge over 30% early bolting rate, which leads to the lignified roots that are useless in medicinal agents. The early bolting and flowering that are leading to serious yield reduction has been one of the most serious problems in the production of A. sinensis for many years. Here, based on previously published research articles, monographs, patents as well as practice experiences, the research progresses on the internal and external factors affecting bolting and flowering of A. sinensis, the pathways regulating bolting and flowering, the mechanism revealing bolting and flowering by biotechnological interventions were summarized, with the aim of providing effective pathways jointly internal and external factors for regulating bolting and flowering, as well as references for further revealing the mechanism of the bolting and flowering of A. sinensis.

ABSTRACT

Purpose@#While numerous epidemiological studies have indicated that omega-3 polyunsaturated fatty acids have anticancer properties in various cancers, the effects and mechanisms of eicosapentaenoic acid (EPA) in ovarian cancer cell growth are poorly understood. @*Materials and Methods@#ES2 ovarian clear cell carcinoma cells and SKOV3 adenocarcinoma cells were treated with palmitic acid or EPA, followed by flow cytometry and cell counting to measure apoptosis and proliferation, respectively. A modified protein lipid overlay assay was used to further verify whether EPA was a ligand of G protein–coupled receptor 30 (GPR30) in ES2 cells. The levels of apoptosis-related genes, phosphorylated AKT, and phosphorylated ERK1/2 were detected to explore the underlying mechanism. Finally, inhibitory effect of EPA on tumor growth via GPR30 was determined in vitro and in vivo. @*Results@#EPA suppressed ES2 ovarian clear cell carcinoma cells growth via GPR30, a novel EPA receptor, by inducing apoptosis. As a ligand of GPR30, EPA activated the GPR30-cAMP– protein kinase A signaling pathway. When GPR30 was suppressed by siRNA or its inhibitor G15, the antiproliferative action of EPA was impaired. Furthermore, EPA inhibited tumor growth by blocking the activation of AKT and ERK. In the mouse xenograft model, EPA decreased tumor volume and weight through GPR30 by blocking tumor cell proliferation. @*Conclusion@#These results confirm that EPA is a tumor suppressor in human ovarian clear cell carcinoma cells and functions through a novel fatty acid receptor, GPR30, indicating a mechanistic linkage between omega-3 fatty acids and cancers.

ABSTRACT

The macroporous microspheres were prepared through suspension polymerization and based on a copolymer of glycidyl methacrylate and ethylene glycol dimethacrylate. The effect of porogen on the microspheres structure was evaluated in terms of pore size and surface area.Porogen contained dichloromethane (δ=9.7 (cal/cm3)1/2) and N-octanol (δ=10.3 (cal/cm3)1/2) which corresponded to a good and poor solvent, respectively. The solubility parameter of porogen was controlled in the range of 9.89-10.09 (cal/cm3)1/2.The pore size of microspheres increased with the difference value of solubility parameter between the polymer and the porogen. On the contrary, the surface area of microspheres decreased in this study. The anion exchange media was prepared through coupling poly(ethylene imine) in the microspheres, and the proteins transport was determined by frontal analysis method. The macroporous microspheres with 257 nm pore size could still afford a high proteins capacity (45.1 mg/mL).These macroporous supports showed a large potential in a rapid separation of proteins.

ABSTRACT

Objective:This paper summarizes the practical problems encountered in the implementation of the tuition-free medical students' policyin rural areas of Sichuan Province, and provides a reference for the implementation of this policy across the province. Methods :Semi-structured interviews were conducted with the principals of the orientation management policies of the health administrative departments and county level and primary health care in-stitutionsin Sichuan, and the focus group interviews were used to understand the dynamics of directed students. Results :The survey results showed that there were limited channels for students to understand the policy ; students' barren understanding of the work of grass-roots doctors ; credit and integrity files played a relatively low role in retaining medical talent students;the willingness of orientation students is higher, but there is a contradiction between applying for graduate students and retaining grassroots in rural areas ; and the relevant departments lacked a rational and efficient management mode for students. Conclusions :In order to ameliorate the construction of rural health human resources, policy administrators should place their emphasis on cultivating the sense of honor and responsibility in the rural-oriented tuition-free medical students'policy, improve the reward and punishment mechanism in policies, optimize the management and assessment process, and promote the orientation of the work among the various policy-participating departments in a more harmonious and orderly manner.

ABSTRACT

Objective: A transferrin and folic acid co-modified Coicis Semen oil-tripterine microemulsion (Tf/FA-CT-MEs) was developed to improve the tumor targeting and thereby enhance the in vitro antitumor efficacy. Methods: FA-PEG 400 was synthesized by classic condensation and together with transferrin as targeting ligands. The structures were characterized by FT-IR and H-NMR. Tf/FA-CT-MEs were prepared by aqueous titration method using Coicis Semen oil as oil phase, followed by anchoring Tf on the surface of prepared particles mentioned-above in aqueous environment. The physiochemical properties and morphology were detected by dynamic light scattering (DLS) and transmission electron microscopy (TEM), respectively. The A549 and MCF-7 cellular uptake of various microemulsions was detected through employing fluorescein isothiocyanate (FITC) as a fluorescence probe. The in vitro antitumor efficacy of the microemulsions against A549 and MCF-7 cells were determined by MTT assay and cell apoptosis kit. Results: FA-PEG 400 was synthesized and characterized as a target ligand. The Tf/FA-CT-MEs with spherical shape had a small droplet size (52.52 ± 0.11) nm, narrow polydispersion index (PDI, 0.124 ± 0.019), and negative Zeta potential (-21.50 ± 1.70) mV, respectively, with good in vitro stability. In in vitro antitumor efficacy, the half proliferation inhibitory concentration (IC50) of Tf/FA-CT-MEs was 0.77 μmol/L on MCF-7 cell model and 0.85 μmol/L on A549 cell model. The cellular uptake results suggested that the fluorescence intensity of MCF-7 and A549 cells was 2782.33 ± 131.77 and 2762.91 ± 23.18 for 4 h, respectively. Besides, (70.60 ± 6.92)% of MCF-7 cells were induced to apoptosis by Tf/FA-CT-MEs. Conclusion: Tf/FA-CT-MEs could improve in vitro targeting and cytotoxicity toward MCF-7 cells. This study suggests that the modification with dul-targeted ligands could be a promising strategy for developing tumor-specific nanomedicines.

ABSTRACT

Based on the clinical symptom characteristics of transient ischemic attack in Chinese and Western medicines, the existing models of transient ischemic attack were summarized and analyzed. Then the advantages and disadvantages of each model, the diagnostic criteria of traditional Chinese and Western medicine and clinical symptoms compliance were analyzed to put forward the evaluation method and improvement method of the corresponding animal models. It was found that there were many modeling methods of transient ischemic attack, but they can not reflect the transience, reversibility, recurrence and other typical characteristics of the disease, with significant differences with clinical symptoms. Moreover, there is lack of reasonable quantitative criteria for the success of the animal model. By combining the existing single factor animal models, a composite animal model that was more closely related to the clinical symptoms of transient ischemic attack was established to replicate an animal model that was more compatible with the characteristics of clinical symptoms. It is the future development directions of the transient ischemic attack animal models to establish reasonable quantitative standards, reflect the causes of Chinese and Western medicine symptoms and improving a series of systematic and complete model evaluation methods.

ABSTRACT

Colorectal cancer (CRC) is one of the most prevalent forms of cancer that shows a high mortality and increasing incidence. There are numerous successful treatment options for CRC, including surgery, chemotherapy, radiotherapy and immunotherapy; however, their side effects and limitations are considerable. Probiotics may be an effective strategy for preventing and inhibiting tumour growth through stimulation of host innate and adaptive immunity. We investigated and compared potential anti-tumour immune responses induced by two isolated Lactobacillus strains, Lactobacillus plantarum A and Lactobacillus rhamnosus b, by pre-inoculating mice with lactobacilli for 14 days. Subsequently, subcutaneous and orthotopic intestinal tumours were generated in the pre-inoculated mice using CT26 murine adenocarcinoma cells and were assessed for response against the tumour. Our results indicated that oral administration with L. plantarum inhibited CT26 cell growth in BALB/c mice and prolonged the survival time of tumour-bearing mice compared with mice administered L. rhamnosus. L. plantarum produced protective immunity against the challenge with CT26 cells by increasing the effector functions of CD8+ and natural killer (NK) cell infiltration into tumour tissue, up-regulation of IFN-γ (but not IL-4 or IL-17) production, and promotion of Th1-type CD4+ T differentiation. Consequently, our results suggest that L. plantarum can enhance the anti-tumour immune response and delay tumour formation.

ABSTRACT

AlM:To observe the relationship between axial length and complications of phacoemulsification with intraocular lens ( lOL) implantation in high axial myopia eyes and normal axis eyes. METHODS: A retrospective review of 843 consecutive patients ( 1 042 eyes ) of cataract extraction with phacoemulsification and lOL implantation in our hospital from February 2012 to February 2013 was performed. The patients were divided into two groups according to the axial length: 853 eyes were in normal axis group ( 21-24mm) and 189 eyes were in high axial myopia group (≥26mm). The two groups were compared regarding surgical complications, such as vitreous loss, posterior capsular rupture, nucleolus drop, and abnormal location of lOL. RESULTS:Age was a risk factor in both groups. There was positive correlation between age and surgical complications, and between axial length and surgical complications, especially for complications with posterior capsular rupture and vitreous loss. CONCLUSlON:As the results illustrate, in this survey, age and high axial lengthare statistically significant risk factors for incidence of complications of phacoemulsification. Anticipation of these complications and also preparation and prophylactic measures may decrease incidence of these complications.
